Spiral thickening in wood is a complex shape which is present in the inner structure of longitudinal cells in some softwood species. Such thickening of cell wall can reach sizes of less than 1 μm and therefore the scanning electron microscopy (SEM) is the suitable imag­ing technique to observe such small structures. On the other hand, focused ion beam scanning electron microsco­py (FIB-SEM) can be used to reveal in detail the microstructure of such speci­mens in the form of 3D reconstructions
